Biological Background: Tyrosine Hydroxylase (TH) Function and Localization
- Tyrosine hydroxylase (TH) catalyzes the rate-limiting step in catecholamine biosynthesis: the conversion of L-tyrosine to L-Dopa, a precursor for dopamine, noradrenaline, and adrenaline. This reaction uses tetrahydrobiopterin and molecular oxygen; it can also hydroxylate phenylalanine and tryptophan with lower specificity. - In immunofluorescence (IF/ICC), permeabilization may be required to access cytoplasmic and nuclear pools of TH. Validate staining in relevant cell lines (e.g., SH-SY5Y neuroblastoma) or primary neurons.
- Since the immunogen is a synthetic peptide, blocking with the immunizing peptide can help confirm antibody specificity.
- For ELISA, optimize coating and detection conditions empirically. The antibody detects human, mouse, and rat TH, enabling cross-species assay development.
