Biological Background: Tyrosine Hydroxylase Function and Localization
- Tyrosine Hydroxylase (TH, TYH) catalyzes the conversion of L-tyrosine to L-dihydroxyphenylalanine (L-Dopa), the rate-limiting step in catecholamine (dopamine, noradrenaline, adrenaline) biosynthesis.

- TH is predominantly expressed in the brain and adrenal glands.
- The enzyme localizes to the cytoplasm, perinuclear region, nucleus, neuronal axons, and synaptic vesicles.
- TH functions as a phosphoprotein, binds iron, and is associated with dystonia (DYT14, DYT5b) and Parkinson’s disease.
- TH can also hydroxylate phenylalanine and tryptophan with lower efficiency.
- It is involved in the positive regulation of retinal hyaloid vessel regression during postnatal development.
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 411-528) of human TH, suggesting the antibody may recognize all full-length isoforms.
- For Western blotting, a band around 59 kDa is expected; consider using brain or adrenal gland lysates as positive controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), validate fixation and permeabilization conditions, as TH localizes to both cytoplasmic and nuclear compartments.
- Cross-reactivity with mouse and rat is reported; however, verify species-specific reactivity in your experimental system.
- For ELISA applications, consider pairing with a detection antibody recognizing a different epitope to avoid competition.
- This recombinant rabbit monoclonal antibody offers batch-to-batch consistency suitable for reproducible assays.
