Biological Background: Tyrosinase Function and Localization
- Tyrosinase, encoded by the TYR gene, is also known as Monophenol monooxygenase, LB24-AB, SK29-AB, and Tumor rejection antigen AB.
- It is a copper-containing oxidase that catalyzes the rate-limiting step in melanin biosynthesis: the hydroxylation of tyrosine to DOPA (3,4-dihydroxyphenylalanine) (By similarity).
- Additionally, tyrosinase oxidizes DOPA to DOPA-quinone and may oxidize DHI (5,6-dihydroxyindole) to indole-5,6 quinone, playing a central role in pigment production (PubMed:28661582).
Related references:
PMID:28661582 - The protein localizes to the melanosome membrane, consistent with its role in melanin synthesis within melanosomes.
- Tyrosinase is associated with various pigmentation disorders; mutations in TYR are linked to oculocutaneous albinism type 1 (OCA1) and Waardenburg syndrome, and the protein is implicated in deafness.
- Post-translational modifications include glycosylation, and the protein is a single-pass transmembrane helix protein with a signal peptide.
Experimental Guidance and Technical Tips
- The immunogen, a synthetic peptide spanning amino acids 20–340 of human Tyrosinase, may exhibit conservation across human, mouse, and rat given observed cross-reactivity; consider validating in the relevant species’ sample system.
- For Western blotting, a band around 60 kDa is expected; optimize loading controls and antibody dilution in your lysate of interest.
- When using IHC-P, consider appropriate antigen retrieval methods and validate staining in melanocyte-rich tissues or positive control samples.
- For IF-P, fixation and permeabilization conditions may influence detection; test on known positive cells expressing Tyrosinase.
- ELISA performance can be assessed using recombinant Tyrosinase or peptide antigen as a standard; ensure proper coating and blocking conditions.
