Biological Background: PML Function and Localization

  • PML acts as the scaffold protein of PML nuclear bodies (PML-NBs) implicated in tumor suppression, transcriptional regulation, apoptosis, senescence, DNA damage response, and viral defense.   • Isoform PML-4 crucially regulates apoptosis and growth suppression: activates RB1, inhibits AKT1 via phosphatases, negatively modulates the PI3K pathway (inhibiting MTOR, activating PTEN), and positively regulates p53 by promoting acetylation/phosphorylation and blocking MDM2-mediated degradation.
