Biological Background: PML Function and Localization
- PML acts as the scaffold protein of PML nuclear bodies (PML-NBs) implicated in tumor suppression, transcriptional regulation, apoptosis, senescence, DNA damage response, and viral defense. - Isoform PML-4 crucially regulates apoptosis and growth suppression: activates RB1, inhibits AKT1 via phosphatases, negatively modulates the PI3K pathway (inhibiting MTOR, activating PTEN), and positively regulates p53 by promoting acetylation/phosphorylation and blocking MDM2-mediated degradation.
- PML-4 functions as a transcriptional repressor of TBX2 during cellular senescence, requires a functional RBL2/E2F4 complex, regulates DNA double-strand break repair via WRN interaction, negatively controls telomerase by binding TERT, and modulates PER2 nuclear localization and circadian rhythm.
