  • TWIST1, also known as H-twist, is a class A basic helix-loop-helix (bHLH) transcription factor encoded by the TWIST1 gene.
  • It functions as a transcriptional regulator, inhibiting myogenesis by sequestering E proteins, blocking MEF2 transactivation, and disrupting MYOD1 DNA-binding through direct interaction.
  • TWIST1 represses expression of pro-inflammatory cytokines such as TNFA and IL1B, and regulates cranial suture patterning and fusion.
  • Depending on its dimerization partner, TWIST1 can either activate or repress target genes; for example, homodimers induce FGFR2 and POSTN, while heterodimers repress these and induce THBS1.
  • Heterodimer formation with E proteins is required for osteoblast differentiation, highlighting its role in bone development.
  • In the nucleus, TWIST1 also represses the circadian transcriptional activator complex NPAS2-BMAL1, linking it to biological rhythm regulation.
