Biological Background: Twist Function and Localization

  • TWIST1 (Twist-related protein 1) is a class A basic helix-loop-helix (bHLH) transcription factor, also known as bHLHa38 or H-twist.
  • Functions as a transcriptional regulator, playing a critical role in craniofacial development by regulating cranial suture patterning and fusion.
  • Inhibits myogenesis through multiple mechanisms, including sequestration of E proteins, inhibition of MEF2 transactivation, and physical interaction with MYOD1 to block its DNA binding.
  • Represses expression of pro-inflammatory cytokines such as TNFA and IL1B, and also represses the activity of the circadian NPAS2-BMAL1 transcriptional activator complex (by similarity).
  • Dimerization status modulates gene expression: homodimers induce FGFR2 and POSTN, while heterodimers with E proteins repress these genes and induce THBS1; heterodimerization is required for osteoblast differentiation.
  • Localized to the nucleus and predominantly expressed in a subset of mesodermal cells.
