Biological Background: TWF2 Function and Localization

  • Twinfilin-2 (also known as A6-related protein, PTK9L, MSTP011) is an actin-binding protein encoded by the TWF2 gene in humans.
  • It inhibits actin polymerization by sequestering G-actin and capping the barbed ends of actin filaments, thereby regulating cytoskeletal dynamics and cell motility.
  • Plays an important role in clathrin-mediated endocytosis and the distribution of endocytic organelles.
  • Involved in regulating the mature length of the middle and short rows of stereocilia in hair cells.
  • Localizes to the cytoplasm, with enrichment in the perinuclear region and association with the cytoskeleton. Also found at cell projections, specifically stereocilia.
  • Ubiquitously expressed at the protein level.
  • Contains actin-binding repeats and is subject to post-translational modifications including acetylation and phosphorylation.

  • For Western blotting, consider using whole cell lysates from human, mouse, or rat cells; the expected band is around 40 kDa. Optimize blocking and antibody dilution conditions for your specific sample system.
