Biological Background: TTF1 Function and Localization
- TTF1 is a multifunctional nucleolar protein that acts as a transcription termination factor for RNA polymerase I (Pol I) on ribosomal DNA (rDNA).
- It mediates replication fork arrest at the rDNA termination site, thereby contributing to DNA replication control.
- TTF1 plays a dual role in rDNA regulation, being involved in both transcriptional activation and silencing of ribosomal genes.
- Interaction with BAZ2A/TIP5 (part of the NoRC complex) can recover TTF1’s DNA-binding activity, linking it to heterochromatin formation and gene silencing.
- The protein localizes predominantly to the nucleus, with enrichment in the nucleolus and presence in the nucleoplasm.
- TTF1 is subject to post-translational modifications including phosphorylation and ubiquitin-like conjugation, and contains DNA-binding domains and repeat regions.
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 806–905) of human TTF1; consider that the antibody may recognize epitopes in this region and may detect full-length TTF1 as well as any C-terminal isoforms.
- For immunofluorescence (IF/ICC), optimize fixation (e.g., 4% PFA) and permeabilization (e.g., 0.1% Triton X-100) to ensure adequate nuclear and nucleolar staining.
- In ELISA, validate the antibody’s performance with recombinant TTF1 protein or cell lysates from appropriate human cell lines to confirm specificity and sensitivity.
- As a polyclonal antibody, it may offer higher sensitivity due to recognition of multiple epitopes; however, lot-to-lot variability should be assessed for critical applications.
