Biological Background: TTC5 Function and Localization
- TTC5, also known as Strap (Stress-responsive activator of p300), is a tetratricopeptide repeat (TPR) protein that functions as a cofactor in diverse cellular processes including actin dynamics, autophagy, chromatin regulation, and DNA repair. Related references: PMID:18451878 PMID:31727855
- Under non-stress conditions, cytoplasmic TTC5 binds JMY, inhibiting JMY’s actin nucleation and Arp2/3 activation, and also suppresses autophagy by blocking JMY’s interaction with MAP1LC3B.
- In response to DNA damage, TTC5 is phosphorylated by ATM and CHEK2 and accumulates in the nucleus, where it facilitates assembly of a p53 coactivator complex with JMY and p300, enhancing p53-dependent transcription and apoptosis.
- TTC5 participates in tubulin autoregulation by associating with ribosomes and nascent tubulin polypeptides, triggering tubulin mRNA degradation when soluble tubulin is in excess. Related references: PMID:31727855
