Biological Background: TTBK2 Function and Localization

  • TTBK2 (tau-tubulin kinase 2) is a serine/threonine protein kinase encoded by the TTBK2 gene; alternative splicing generates multiple isoforms.
  • Acts as a key regulator of ciliogenesis: binds to the distal end of the basal body and promotes removal of CCP110, capping the mother centriole, allowing recruitment of IFT proteins to build the ciliary axoneme (PubMed:23141541). Additionally, TTBK2 phosphorylates MPHOSPH9, targeting it for degradation, which facilitates removal of the CP110-CEP97 complex from mother centrioles (PubMed:30375385). Related references: PMID:23141541 PMID:30375385
  • Phosphorylates tau protein on serines in vitro, linking TTBK2 to tau pathology (PubMed:23141541). Related references: PMID:23141541
  • Localizes to multiple subcellular compartments, including the cilium basal body, centrosome, centriole, cytosol, and nucleus.
  • Classified as a phosphoprotein and is associated with spinocerebellar ataxia type 11 (SCA11), a neurodegenerative disorder.
  • Keywords include ATP-binding, nucleotide-binding, kinase, transferase, and cilium biogenesis/degradation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1185-1244 near the C-terminus; the antibody may detect full-length TTBK2 and certain isoforms.
  • For Western blotting, the predicted molecular weight is 137 kDa; consider that post-translational modifications (e.g., phosphorylation) may shift migration.
  • When using for immunohistochemistry (IHC-P), optimize antigen retrieval conditions for your specific tissue and fixation.
  • ELISA applications may be employed with recombinant or enriched endogenous protein samples; validate assay conditions.
  • Cross-reactivity with rat is reported; validate in the relevant species prior to experimental use.
