Biological Background: TSPYL2 Function and Localization
-
TSPYL2 (Testis-specific Y-encoded-like protein 2), also known as CDA1, DENTT, or TSPX, is a protein encoded by the TSPYL2 gene in humans.
-
It participates in the CASK/TBR1/TSPYL2 transcriptional complex, modulating gene expression in response to neuronal synaptic activity by facilitating nucleosome assembly.
-
TSPYL2 may inhibit cell proliferation by inducing p53-dependent CDKN1A expression, highlighting a role in cell cycle regulation.
-
The protein localizes to both the nucleus and cytoplasm, consistent with its involvement in transcriptional regulation.
-
Ubiquitously expressed, TSPYL2 shows highest levels in brain, testis, and heart, with lowest levels in liver and pancreas.
-
Post-translational modifications include phosphorylation and ubiquitination (isopeptide bond, Ubl conjugation), as indicated by UniProt keywords, and it functions as a chromatin regulator.
Experimental Guidance and Technical Tips
-
When performing Western blotting, expect a band at approximately 79 kDa; due to the polyclonal nature of the antibody, additional bands from isoforms or degradation may appear—validate in your specific sample system.
-
For ELISA applications, consider the target's presence in the sample type and optimize coating and detection conditions accordingly.
-
The immunogen (amino acids 240-460) covers the C-terminal region; therefore, this antibody recognizes the C-terminal part of TSPYL2, which may be useful in probing specific domain functions.
-
Cross-reactivity with mouse and rat is indicated; however, verify reactivity in your own experimental context, especially if working with tissues with varying expression levels.
