Biological Background: TSPO Function and Localization

  • TSPO, also known as translocator protein, mitochondrial benzodiazepine receptor, PKBS, or peripheral-type benzodiazepine receptor (PBR), is encoded by the TSPO gene in humans. The protein undergoes alternative splicing, producing isoforms that may vary in expression and function.
  • It is a multi-pass transmembrane protein located on the mitochondrion membrane, consistent with its role in mitochondrial transport processes.
  • TSPO can bind protoporphyrin IX and may be involved in the transport of porphyrins and heme, suggesting a role in heme biosynthesis or metabolism.
  • It promotes the transport of cholesterol across mitochondrial membranes and may participate in lipid metabolism, although its exact physiological role remains controversial. Related references: PMID:24814875
  • Surprisingly, TSPO is apparently not required for steroid hormone biosynthesis, challenging its long-proposed function in steroidogenesis.
  • The protein was initially characterized as a peripheral-type benzodiazepine receptor and also binds isoquinoline carboxamides, implicating it in neuropharmacological pathways. Related references: PMID:1847678
  • TSPO is widely expressed but shows highest levels in tissues that synthesize steroids, such as adrenal cortex, gonads, and brain, aligning with its proposed role in cholesterol transport for steroidogenesis.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein corresponding to the N-terminal region (amino acids 1-50) of human TSPO, which may provide specificity for the target.
  • For Western blot applications, consider using reducing or non-reducing conditions as needed; the predicted molecular weight is 19 kDa, but post-translational modifications or dimerization may affect migration.
  • In IF/ICC, fixation and permeabilization protocols should be optimized for mitochondrial membrane proteins; confirm localization with mitochondrial markers.
  • When performing ELISA, titrate the antibody and validate with a recombinant TSPO protein or cell lysates to establish linear range and sensitivity.
  • Cross-reactivity with mouse TSPO is reported, making this antibody suitable for rodent studies; validate in the appropriate tissue type where TSPO is highly expressed.
