Biological Background: TSPAN4/NAG-2 Function and Localization

  • Tetraspanin-4 (TSPAN4), also known as NAG-2 and TM4SF7, is a member of the tetraspanin superfamily, characterized by four transmembrane domains.
  • It acts as a structural component of specialized membrane microdomains known as tetraspanin-enriched microdomains (TERMs), facilitating receptor clustering and signaling.
  • TSPAN4 plays an essential role in migrasome formation and migration on retracting fibers at the rear of migrating cells.
    Related references: PMID:31371828
  • It functions as a membrane curvature sensor and stabilizes the migrasome structure during late-stage biogenesis.
    Related references: PMID:36252000, PMID:36823145
  • TSPAN4 may also regulate the histamine H4 receptor (HRH4) without affecting histamine binding or signaling.
    Related references: PMID:34439793
  • Subcellularly, TSPAN4 is localized to the cell membrane.
  • It is expressed in multiple tissues but is absent in brain, lymphoid cells, and platelets.
  • Post-translationally, TSPAN4 is a glycoprotein with a calculated molecular weight of approximately 26 kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 100–200 of human TSPAN4, a region within the large extracellular loop. This design may favor detection of the target in both native and denaturing conditions.
  • For Western blotting, consider optimizing antibody concentration and using appropriate loading controls. The predicted molecular weight is ~26 kDa; however, post-translational modifications may affect migration.
  • In ELISA applications, direct or indirect formats can be employed; titration is recommended to determine optimal working dilutions.
  • Cross-reactivity with mouse and rat TSPAN4 has been noted; however, validation in the relevant sample system is advised.
