Biological Background: TSPAN33 Function and Localization

  • TSPAN33 is a member of the tetraspanin family and belongs to the TspanC8 subgroup, which comprises six members that interact with the transmembrane metalloprotease ADAM10. This interaction is essential for ADAM10 exit from the endoplasmic reticulum, its enzymatic maturation, trafficking to the cell surface, and determination of substrate specificity.   • Plays a critical role in normal erythropoiesis and the differentiation of erythroid progenitors (By similarity).

  • Negatively regulates ligand-induced Notch activity, likely by modulating ADAM10 function. Related references: PMID:26686862

  • Mediates the docking of ADAM10 to the zonula adherens by simultaneously interacting with ADAM10 and, in a PDZD11-dependent manner, with the adherens junction protein PLEKHA7. Related references: PMID:30463011
