Biological Background: Tspan18 Function and Localization
- Tetraspanin-18 (Tspan18), also known by aliases 6720430O15 and 2610042G18Rik, is a member of the tetraspanin family of transmembrane proteins.
- It plays a role in the cell surface localization of ORAI1, a pore-forming subunit of store-operated calcium channels, thereby participating in the regulation of Ca2+ signaling.
- Tspan18 is implicated in the release of von Willebrand factor (VWF) from endothelial cells in response to inflammatory stimuli.
- Localized to the membrane, Tspan18 is a multi-pass membrane protein with several transmembrane helices.

Experimental Guidance and Technical Tips
- This polyclonal antibody is raised against a synthetic peptide corresponding to amino acids 149–248 of mouse Tspan18. Peptide blocking experiments can be used to confirm specificity in your assay system.
- For Western blot, the predicted molecular weight of Tspan18 is approximately 28 kDa (UniProt: 27.8 kDa). A band migrating between 25–30 kDa is expected; consider using a protein ladder with appropriate resolution.
- For ELISA, the antibody may be used as a capture or detection reagent. Optimal coating concentrations and detection dilutions should be determined empirically.
- Given its membrane localization, enrichment of membrane fractions or immunoprecipitation prior to WB may improve detection.
- Cross-reactivity has been observed with mouse and rat Tspan18. Due to high sequence conservation, reactivity with other species (e.g., human) is plausible but should be validated in the target sample.
