Biological Background: TSNAX Function and Localization
- TSNAX, also known as Translin-associated protein X or TRAX, is a protein that functions in combination with translin (TSN).
- The TSNAX/TSN complex acts as an endonuclease involved in the activation of the RNA-induced silencing complex (RISC), playing a role in RNA interference pathways.
- TSNAX may have a role in spermatogenesis, as suggested by its association with developmental processes.
- Subcellular localization studies indicate TSNAX is found in the cytoplasm, particularly in the perinuclear region, as well as in the Golgi apparatus and nucleus.
- The protein contains DNA-binding domains and requires magnesium as a cofactor, consistent with its nuclease activity.
- Post-translational modifications include isopeptide bond formation and ubiquitination (Ubl conjugation), which may regulate its function or stability.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant protein covering amino acids 1-290 of human TSNAX; this full‐length fragment may generate polyclonal antibodies recognizing multiple epitopes, which can enhance detection sensitivity but may require careful blocking to minimize non‐specific binding.
- For Western blot (WB), expect a band at approximately 33 kDa in lysates from human, mouse, or rat cells expressing TSNAX. Consider using a positive control such as HeLa or testis tissue lysate, where TSNAX is known to be expressed.
- For ELISA, the recombinant immunogen can serve as a positive coating antigen; validate assay conditions (coating concentration, blocking, and secondary antibody dilution) using a known TSNAX-containing sample.
- Cross‐reactivity with mouse and rat has been reported; however, sequence homology should be checked if using for non‐human species to ensure epitope conservation. Verify in the relevant sample system before quantitative applications.
