Biological Background: TSHZ3 Function and Localization
- TSHZ3 (Teashirt homolog 3), also known as Zinc finger protein 537, is a transcriptional regulator encoded by the TSHZ3 gene, belonging to the teashirt zinc finger family.
- Functions as a transcriptional repressor by associating with the chromatin near the CASP4 transcriptional start site(s) and recruiting histone deacetylases HDAC1 and HDAC2 to inhibit CASP4 expression. Related references: PMID:19343227
- Plays a critical role in neurodevelopment: it regulates the differentiation and maintenance of neurons controlling respiratory rhythm and airflow, including nucleus ambiguus motoneurons, and is essential for postnatal survival.
- Involved in smooth muscle differentiation in the proximal ureter by up-regulating myocardin, thereby directing the expression of smooth muscle cell-specific genes (By similarity).
- Subcellular localization encompasses the nucleus, and it is also detected in cell projections, specifically growth cones, suggesting roles in neurite outgrowth or guidance.
- Tissue expression is prominent in the brain, particularly in the fetal neocortex; notably, its levels are strongly reduced in post-mortem brains of elderly subjects with Alzheimer disease, implying a potential link to neurodegeneration.
- Structurally, TSHZ3 contains a homeobox domain, multiple zinc finger motifs, a coiled-coil region, and undergoes phosphorylation, consistent with its roles in DNA binding, protein interactions, and signal-responsive regulation.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant protein covering amino acids 791–890 of human TSHZ3, a C-terminal region. This region may contain a unique epitope; verify its accessibility in the experimental context.
- For Western blotting, a protein band at ~119 kDa is expected. Use standard high-molecular-weight transfer conditions (e.g., 10% SDS-PAGE, wet transfer) and consider optimizing blocking and antibody dilutions (e.g., 1:500–1:2000) for your sample.
- For ELISA applications, test direct or indirect formats with coating antigen at 1–10 µg/mL and primary antibody dilutions between 1:1000 and 1:5000. Validate specificity using positive (e.g., TSHZ3-overexpressing lysates) and negative controls.
- As the antibody shows cross-reactivity with mouse TSHZ3, it can be used for comparative studies; however, validate in the relevant mouse tissue or cell line to ensure consistent performance.
