Biological Background: TSG101/VPS23 Function and Localization
- TSG101 (Tumor susceptibility gene 101 protein), also known as ESCRT-I complex subunit TSG101, is a component of the ESCRT-I complex, which regulates vesicular trafficking by binding ubiquitinated cargo proteins and sorting them into multivesicular bodies (MVBs).
- Acts as a linker between ESCRT-0 and ESCRT-I complexes, essential for the MVB pathway and lysosomal degradation of transmembrane proteins.
- Required for completion of cytokinesis; interacts with CEP55 at the midbody ring, playing a role in cell division.
- Involved in the budding of many enveloped viruses by interacting with viral late-budding motifs (P-[ST]-A-P), crucial for viral particle release from host cells.
- Required for the exosomal release of SDCBP, CD63, and syndecan. Related references: PMID:22660413
- May also participate in the extracellular release of microvesicles that differ from exosomes. Related references: PMID:22315426
- Subcellular localization includes cytoplasm, early and late endosome membranes, centrosome, midbody ring, and nucleus.
- Tissue expression is broad: heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas. Post-translational modifications include phosphorylation, acetylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 291–390 of human TSG101, located in the C-terminal region. This region is relatively conserved; cross-reactivity with mouse and rat TSG101 has been observed in manufacturer testing. Validate specificity in your model system.
- Predicted molecular weight: ~44 kDa. In WB, a band near 44 kDa is expected; additional bands may appear due to post-translational modifications or isoforms. Use appropriate loading controls.
- For IF/ICC, the antibody may detect TSG101 at various subcellular locations (endosomes, midbody, nucleus). Optimize fixation and permeabilization conditions for your specific cell type and target compartment.
- In ELISA, the antibody can be used for peptide or protein detection. Titrate antibody concentration and consider using a blocking buffer to reduce background.
