- TSG101 (Tumor susceptibility gene 101 protein), also known as VPS23, is a core component of the ESCRT-I complex, which regulates vesicular trafficking by binding ubiquitinated cargo and mediating sorting into multivesicular bodies (MVBs).
- It plays a critical role in cytokinesis by interacting with CEP55 at the midbody, ensuring proper cell division.
- TSG101 acts as a negative growth regulator and may be involved in cell growth and differentiation.
- The protein is hijacked by many enveloped viruses (e.g., HIV-1) via interaction with late-budding motifs (P-[ST]-A-P) to facilitate viral particle budding and release.

- ELISA may be used for peptide-specific detection; validate assay conditions with the immunizing peptide as a positive control.
