Biological Background: TSC1 Function and Localization
- Hamartin (TSC1) is a core subunit of the TSC-TBC complex that negatively regulates the mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway. Related references: PMID:12172553 PMID:12271141 PMID:12906785
- The TSC-TBC complex functions as a GTPase-activating protein (GAP) for RHEB, directly inhibiting mTORC1 kinase activity. Related references: PMID:12906785 PMID:15340059 PMID:24529379
- Under nutrient-deprived conditions, TSC1/TSC2 suppresses mTORC1, blocking phosphorylation of S6 kinases and 4E-BP1; nutrient signals relieve this inhibition. Related references: PMID:12271141 PMID:24529379 PMID:28215400
- TSC1 stabilizes TSC2 by preventing its self-aggregation and recruits TSC2 to HSP90AA1, protecting it from ubiquitination and degradation. Related references: PMID:10585443 PMID:28215400 PMID:29127155 PMID:16464865
- Acts as a tumor suppressor; loss-of-function mutations in TSC1 are associated with tuberous sclerosis complex. Related references: PMID:9242607
- Subcellular localization includes the lysosome membrane, cytoplasm, and cytosol, consistent with its role in nutrient sensing.
- Highly expressed in skeletal muscle, heart, brain, placenta, pancreas, lung, liver, and kidney. Related references: PMID:9242607
- Also acts as a co-chaperone for HSP90AA1, modulating ATP binding and client protein folding, and competes with AHSA1. Related references: PMID:29127155
Experimental Guidance and Technical Tips
- The immunogen corresponds to a conserved region (AA 600-700) of human TSC1; observed cross-reactivity with mouse and rat suggests suitability for multi-species studies.
- For Western blot (WB), TSC1 migrates at approximately 130 kDa; consider using a low-percentage gel and optimized transfer conditions for high-molecular-weight proteins.
- For IHC-P and IF/ICC, validate fixation and antigen retrieval protocols in the relevant tissue or cell system to ensure specific detection.
- ELISA applications may require titration of the antibody using the immunizing peptide or recombinant TSC1 protein as a standard.
