Biological Background: Tryptophanyl-tRNA synthetase 1 Function and Localization
- Tryptophanyl-tRNA synthetase 1, also designated tryptophan--tRNA ligase, cytoplasmic, is encoded by the WARS1 gene and commonly referred to as TrpRS, IFP53, or WARS.
- It catalyzes the ATP-dependent ligation of tryptophan to its cognate tRNA, a critical step in cytoplasmic protein synthesis.
- Alternative splicing generates isoforms with distinct functions; one isoform lacks tRNA aminoacylation activity but acts as an angiostatic factor by inhibiting endothelial cell responses to fluid shear stress and modulating ERK/Akt/eNOS signaling pathways. - The protein localizes primarily within the cytoplasm.
- It belongs to the class-I aminoacyl-tRNA synthetase family and contains conserved ATP-binding and nucleotide-binding motifs.
- Post-translational modifications include phosphorylation, and the protein is implicated in neurological disorders such as neurodegeneration and neuropathy.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1-270) of human TrpRS. For Western blot, expect a band near 53 kDa under reducing conditions; multiple bands may appear due to alternative splicing or post-translational modifications.
- In ELISA applications, consider that the polyclonal nature may capture both full-length and angiostatic isoforms; validate specificity with recombinant isoform controls.
- When using mouse or rat samples, verify cross-reactivity in the target tissue and consider that sequence homology in the immunogen region may influence detection efficiency.
