Biological Background: TRPM8 Function and Localization

  • TRPM8 (Transient Receptor Potential Cation Channel Subfamily M Member 8), also known as LTrpC-6 and Trp-p8, is a transmembrane ion channel of the TRPM subfamily.
  • It functions as a cold sensor, activated by cool temperatures (<23–28°C) and chemical ligands such as menthol and icilin, leading to non-selective cation influx (Na⁺, K⁺, Ca²⁺) with higher Ca²⁺ permeability. Related references: PMID:15306801 PMID:15852009 PMID:16174775
  • Channel activation by cold or ligands shifts the voltage dependence of TRPM8 towards physiological membrane potentials, facilitating opening. Related references: PMID:15306801
  • TRPM8 may contribute to prostate cancer cell migration, suggesting a role in tumor biology. Related references: PMID:16174775 PMID:25559186
  • A regulatory mechanism negatively modulates menthol- and cold-induced channel activity by stabilizing the closed state of the channel.
  • The protein localizes to the cell membrane, membrane rafts, and endoplasmic reticulum membrane, consistent with its ion channel and signaling functions.
  • TRPM8 is primarily expressed in prostate tissue and is also found in prostate tumors and other primary tumors (colon, lung, breast, skin), highlighting its potential as a diagnostic or therapeutic target.

Experimental Guidance and Technical Tips

  • The immunogen (synthetic peptide from the cytoplasmic region, aa 900–1000) is expected to recognize the native protein; however, experimental validation in the relevant sample system is advised.
  • For Western blot, due to the large predicted size (128 kDa), consider using a gradient gel and optimized transfer conditions for high-molecular-weight proteins.
  • When performing ELISA, titrate the antibody and include appropriate positive and negative controls to confirm specific binding in your assay format.
