Biological Background: TRPM4 Function and Localization

  • TRPM4, also known as melastatin-4 or LTrpC4, is a member of the transient receptor potential (TRP) channel family and functions as a calcium-activated non-selective cation channel.

  • TRPM4 plays essential roles in diverse tissues: it contributes to pacemaking activity in cardiomyocytes, insulin secretion in pancreatic beta-cells, myogenic constriction of cerebral arteries, and neuronal function in entorhinal cortex and sensory neurons.
  • In the immune system, it regulates T-cell activation by modulating Ca2+ oscillations required for NFAT-dependent IL2 production, and differentially controls T-helper 1 (Th1) and Th2 cell motility and cytokine production.

  • The protein localizes to the cell membrane, endoplasmic reticulum, and Golgi apparatus, and is subject to post-translational modifications including glycosylation, phosphorylation, and ubiquitination.
