Biological Background: TRPC6 Function and Localization
- TRPC6 (Transient receptor potential cation channel subfamily C member 6) is a member of the short transient receptor potential channel (TRPC) subfamily and exists in multiple isoforms due to alternative splicing.
- It functions as a non-selective, calcium-permeable cation channel that mediates calcium influx in response to various stimuli. Related references: PMID:19936226 PMID:23291369 PMID:26892346
- TRPC6 is directly activated by diacylglycerol (DAG) produced via phospholipase C (PLC)-mediated hydrolysis of phosphatidylinositides downstream of Gq-coupled receptors or receptor tyrosine kinases. Related references: PMID:26892346
- Unlike some TRP channels, TRPC6 is not activated by depletion of intracellular calcium stores. Related references: PMID:9930701
- In the retina, TRPC6 contributes to light-induced depolarization of intrinsically photosensitive retinal ganglion cells (ipRGCs) by mediating melanopsin-driven Gq-PLC signaling, likely as part of heteromeric TRPC6-TRPC7 channels.
- TRPC6 localizes to the cell membrane and is highly expressed in placenta, lung, spleen, ovary, small intestine, and podocytes of the kidney glomerular slit diaphragm.
- Mutations in TRPC6 are associated with focal segmental glomerulosclerosis (FSGS2), highlighting its critical role in podocyte function and renal filtration.
- Post-translational modifications include glycosylation and phosphorylation, and the protein contains ankyrin repeats and six transmembrane helices.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide within the cytoplasmic domain (amino acids 466–565), which may be conserved across species, supporting broad reactivity.
- For Western blot (WB): The predicted molecular weight is ~106 kDa; consider using reducing conditions and validate band specificity in relevant cell lysates (e.g., kidney, lung).
- For immunohistochemistry (IHC-P): Antigen retrieval methods may be required; validate staining in tissues known to express TRPC6 (e.g., placenta, podocytes).
- For ELISA: The antibody can be used as a capture or detection reagent; confirm linearity and sensitivity with recombinant TRPC6 or relevant samples.
