Biological Background: TRPC1 Function and Localization
- TRPC1 (Transient receptor potential channel 1), also known as TRP-1, is a member of the transient receptor potential (TRP) ion channel family.
- It forms a receptor-activated, non-selective calcium-permeable cation channel, and can also be activated by intracellular calcium store depletion. Related references: PMID:11139478 PMID:15016832 PMID:39478185
- TRPC1 can assemble into heteromeric channels with TRPC4 or TRPC5, which exhibit reduced calcium permeability compared to the homomeric TRPC4 or TRPC5 channel. Related references: PMID:39478185
- The channel is also permeable to monovalent ions such as sodium, lithium, and cesium. Related references: PMID:39478185
- TRPC1 is ubiquitously expressed in human tissues and localizes to the cell membrane.
- The protein contains multiple ankyrin repeats, transmembrane helices, and undergoes post-translational modifications including phosphorylation, disulfide bond formation, and zinc binding.
- Alternative splicing generates multiple isoforms of TRPC1.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody is a synthetic peptide spanning amino acids 400–500 of human TRPC1, targeting an intracellular region. Consider this when designing epitope-specific detection strategies.
- For Western blotting, the predicted molecular weight of TRPC1 is 88–91 kDa. Validate the observed band size in appropriate positive control lysates.
- In ELISA applications, confirm optimal antibody dilution and coating conditions using recombinant TRPC1 protein or cell lysates expressing the target.
- Cross-reactivity with mouse and rat TRPC1 has been observed; however, always verify specificity in your experimental system with appropriate controls.
