Biological Background: TROVE2/SSA2 (RO60) Function and Localization
- Also known as 60 kDa SS-A/Ro ribonucleoprotein, Ro60 autoantigen, and Sjoegren syndrome antigen A2, TROVE2 is a target of autoantibodies in systemic lupus erythematosus and Sjögren's syndrome.
- Functions as an RNA-binding protein that recognizes misfolded non-coding RNAs, pre-5S rRNA, and Y RNAs, and also binds endogenous Alu retroelements induced by type I interferon, modulating inflammatory cytokine secretion. Related references: PMID:18056422, PMID:26382853
- Plays a role in regulating the expression of Alu retroelements and inflammatory genes, linking it to innate immune responses. Related references: PMID:26382853
- Primarily localized to the cytoplasm, consistent with its function in cytoplasmic RNA surveillance.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-260) of human TROVE2; thus, the polyclonal antibody may recognize epitopes in this region, suitable for both native and denaturing conditions.
- For Western blotting, the expected molecular weight is approximately 61 kDa; consider using whole-cell lysates from human, mouse, or rat tissues and validate with appropriate controls.
- In ELISA, optimize the coating antigen concentration and antibody dilution to achieve desired sensitivity; note that the polyclonal nature may require careful titration.
- Cross-reactivity with human, mouse, and rat has been indicated; verify reactivity in your specific sample system if using other species.
