Biological Background: TRNT1 Function and Localization
- TRNT1 is a nucleotidyltransferase that catalyzes the addition and repair of the essential 3'-terminal CCA sequence in tRNAs, which is required for amino acid attachment, using CTP and ATP as substrates. Related references: PMID:11504732, PMID:25193871, PMID:25640237
- It mediates tRNA repair and recycling downstream of ribosome-associated quality control (RQC) by adding the CCA sequence after ANKZF1 cleavage and ELAC1 repair. Related references: PMID:31011209
- TRNT1 also functions in tRNA surveillance by adding a tandem CCACCA sequence to the 3' terminus of unstable tRNAs, marking them for rapid degradation, while stable tRNAs receive only a single CCA. Related references: PMID:22076379, PMID:25640237
- An isoform exists that adds only two C residues (CC-) to the 3' terminus instead of a complete CCA end.
