Biological Background: TRNAU1AP Function and Localization
- TRNAU1AP (tRNA selenocysteine 1-associated protein 1), also designated SECp43 or TRSPAP1, is encoded by the TRNAU1AP gene in humans. The protein is subject to alternative splicing, generating multiple isoforms (Keywords: Alternative splicing; Reference proteome).
- TRNAU1AP plays a crucial role in the biosynthesis of selenocysteine (Sec) and its cotranslational incorporation into selenoproteins. It participates in early steps of Sec biosynthesis and later steps of tRNA(Sec) charging and recruitment to the ribosome. Related references: PMID:16508009
- The protein stabilizes a ternary complex composed of SECISBP2, EEFSEC (SelB), and tRNA(Sec), thereby facilitating efficient decoding of UGA as selenocysteine. Related references: PMID:16508009
- TRNAU1AP may also be involved in the methylation of tRNA(Sec), a modification important for tRNA stability and function. Its RNA-binding capacity is supported by the presence of RNA-binding domains (Keywords: RNA-binding). Related references: PMID:16508009
- Subcellularly, TRNAU1AP localizes to both the nucleus and cytoplasm (SUBCELLULAR LOCATION: Nucleus, Cytoplasm), consistent with its roles in tRNA processing and translation.
- As a reference proteome member with available 3D structural data (Keywords: 3D-structure; Reference proteome), TRNAU1AP represents a well-characterized target for studying selenoprotein synthesis.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1–287), which may target conserved epitopes, likely contributing to cross-reactivity between human and mouse.
- For Western blot, the predicted molecular weight of TRNAU1AP is ~32 kDa; consider validating the band size in endogenous samples.
- Being a polyclonal antibody, it may recognize multiple isoforms; be aware of potential multiple bands in WB.
- For ELISA, the antibody can be used for detection of recombinant or native TRNAU1AP; appropriate coating and blocking conditions should be optimized.
