Biological Background: TRMT2B Function and Localization
- TRMT2B (tRNA (uracil-5-)-methyltransferase homolog B) is a mitochondrial methyltransferase that catalyzes the formation of 5-methyluridine at position 54 in tRNAs (m5U54) and at position 429 in 12S rRNA (m5U429), utilizing S-adenosyl-L-methionine as a cofactor (PubMed:31948311, PubMed:34556860). Related references: PMID:31948311, PMID:34556860
- The enzyme localizes to the mitochondrial matrix, consistent with its role in mitochondrial RNA modification.
- Also known as TRM2 homolog B, the protein is encoded by the TRMT2B gene (previously CXorf34) and is subject to alternative splicing, producing multiple isoforms.
- TRMT2B is involved in tRNA processing and rRNA processing, as indicated by UniProt annotation, and does not affect RNA stability or mitochondrial translation (PubMed:31948311). Related references: PMID:31948311
- The protein has a predicted molecular weight of approximately 56 kDa (56476 Da) and contains a mitochondrial transit peptide for import.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1–250 of human TRMT2B, which includes the N-terminal region; this may cover the mitochondrial targeting sequence. When designing complementary antibodies or validating epitope accessibility, consider the potential impact of mitochondrial import processing.
- For Western blotting, a band at ~56 kDa is expected; validate expression in mitochondrial-enriched fractions or whole-cell lysates from human and rat cell lines.
- In immunohistochemistry (IHC-P), antigen retrieval methods may be required; test on positive control tissues with known TRMT2B expression, and optimize primary antibody dilution.
- For ELISA, the antibody can be used as a capture or detection reagent; confirm binding specificity using recombinant TRMT2B protein.
