Biological Background: TRMT2A/HTF9C Function and Localization
- TRMT2A, also known as HTF9C, is a tRNA (uracil-5-)-methyltransferase homolog A that functions as an S-adenosyl-L-methionine-dependent methyltransferase.
- It catalyzes the formation of 5-methyluridine (m5U) primarily at position 54 in cytosolic tRNAs, a key modification for tRNA stability and function. Related references: PMID:31361898 PMID:33799331
- Additionally, TRMT2A mediates m5U modification in certain mRNAs, linking it to epitranscriptomic regulation. Related references: PMID:34123281
- The protein localizes to the cytoplasm/cytosol, consistent with its role in processing cytosolic tRNAs and mRNAs.
- TRMT2A contains a methyltransferase domain and features such as a coiled coil region, and is subject to phosphorylation and alternative splicing, as indicated by proteomics and structural studies.
- As a reference proteome member with RNA-binding activity, TRMT2A is evolutionarily conserved and plays a critical role in tRNA and mRNA maturation.
Experimental Guidance and Technical Tips
- The immunogen, spanning amino acids 1-250 of human TRMT2A, corresponds to the N-terminal region; consider that this polyclonal antibody may recognize multiple epitopes.
- For Western blot (WB), the predicted molecular weight is 69 kDa; validate specificity using appropriate positive and negative controls in your sample system.
- When performing immunohistochemistry (IHC-P), optimize antigen retrieval conditions and confirm staining patterns relative to known TRMT2A expression.
- For ELISA, determine optimal coating concentrations and antibody dilutions empirically to achieve the desired signal-to-noise ratio.
