Biological Background: TrkB Function and Localization

  • TrkB (also known as GP145-TrkB, NTRK2) is a receptor tyrosine kinase and a member of the neurotrophic tyrosine kinase receptor family. Multiple isoforms exist, including the full-length TrkB, TrkB-T1, and TrkB-T-Shc, with TrkB being the predominant form in the nervous system.
  • Upon binding to its ligands—brain-derived neurotrophic factor (BDNF), neurotrophin-4 (NTF4), and with lower affinity neurotrophin-3 (NTF3)—TrkB homodimerizes and undergoes autophosphorylation. This activation triggers several downstream signaling pathways, including the GRB2-Ras-MAPK cascade (regulating neuronal differentiation and neurite outgrowth), the PI3K-AKT cascade (promoting survival and growth), and PLCγ-mediated pathways (influencing synaptic plasticity).   • TrkB plays a critical role in the development and maturation of the central and peripheral nervous systems, regulating neuron survival, proliferation, migration, differentiation, and synapse formation. It contributes to learning and memory by modulating both short-term synaptic function and long-term potentiation.
  • Subcellularly, TrkB is localized at the cell membrane, as well as in early endosomes, axonal and dendritic projections, and the postsynaptic density. A perinuclear cytoplasmic pool is also observed.
  • TrkB expression is abundant in the CNS (cerebral cortex, hippocampus, thalamus, cerebellum, brain stem, spinal cord) and in the PNS (cranial ganglia, dorsal root ganglia). The TrkB-T1 isoform is additionally found in non-neuronal tissues such as pancreas, kidney, and heart.
  • Post-translational modifications include glycosylation, ligand-dependent phosphorylation, and ubiquitination. Mutations in NTRK2 are linked to developmental encephalopathies (e.g., DEE58, EIEE58) and obesity.
  • TrkB suppresses anoikis and mediates neurotrophin-dependent calcium signaling in glial cells, facilitating communication between neurons and glia.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 32–430 of human TrkB, a region within the extracellular domain. This antibody is expected to recognize the full-length TrkB isoform and potentially other isoforms sharing this sequence.
  • For Western blotting, the predicted molecular weight is approximately 92 kDa for the canonical TrkB isoform. Due to glycosylation and alternative splicing, bands of different sizes may appear (e.g., TrkB-T1 at ~95 kDa). Use appropriate positive controls such as neuronal cell lines (e.g., SH-SY5Y, PC12) or brain tissue lysates.
  • The antibody cross-reacts with mouse and rat TrkB, according to the supplier. We recommend verifying reactivity in your specific sample matrix.
