Biological Background: TrkA Function and Localization
- TrkA (NTRK1) is a high affinity receptor for nerve growth factor (NGF), playing a central role in the development and maturation of the central and peripheral nervous systems by regulating neuronal proliferation, differentiation, and survival.

PMID:1281417 PMID:15488758 PMID:17196528 - Multiple isoforms exist: TrkA-I (widely expressed in non-neuronal tissues), TrkA-II (predominantly neuronal), and TrkA-III (specifically in pluripotent neural stem and neural crest progenitors), each with distinct signaling properties.
- Upon NGF binding, TrkA homodimerizes, autophosphorylates, and activates downstream effectors including SHC1, FRS2, and PLCG1, which trigger the Ras-MAPK cascade (cell differentiation), PI3K-AKT pathway (survival), and NF-kappa-B signaling, respectively.
- Isoform TrkA-III is NGF-resistant, constitutively activates AKT1 and NF-kappa-B, antagonizes anti-proliferative NGF-NTRK1 signaling, and promotes angiogenesis and oncogenic activity when overexpressed.
- TrkA localizes to the cell membrane and undergoes ligand-dependent endocytosis, residing in early, late, and recycling endosome membranes, which may sustain or diversify signaling.
- The TrkA receptor is a member of the neurotrophic tyrosine kinase receptor family (also including TrkB/NTRK2 and TrkC/NTRK3); this antibody recognizes a conserved epitope across all three Trk receptors, enabling pan-Trk detection.
- Key functional annotations include: ATP-binding, tyrosine-protein kinase, immunoglobulin domain, leucine-rich repeat, transmembrane helix, glycoprotein, phosphoprotein, and Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the intracellular region of human NTRK2 (amino acids 722–822), which is conserved across TrkA, TrkB, and TrkC, accounting for pan-Trk reactivity.
- For Western blotting, expect a band around 140 kDa (glycosylated mature form); reducing conditions are typical. Multiple bands or a doublet may appear due to differential glycosylation or cleavage products; validate in your sample system with appropriate controls.
- For immunohistochemistry (paraffin), antigen retrieval (heat-mediated, citrate buffer pH 6.0 or equivalent) is often required; optimize detection based on tissue type and expression levels.
- For ELISA, the antibody can serve as a capture or detection reagent; consider pairing with a different Trk-specific monoclonal antibody or a pan-Trk polyclonal for sandwich assays.
- The antibody is rabbit monoclonal, ensuring high specificity and lot-to-lot consistency; recommended starting dilutions should be determined empirically for each assay.
