Biological Background: TFG Function and Localization
- Protein TFG (TRK-fused gene protein) is a ubiquitously expressed protein encoded by the TFG gene, also known as TF6, HMSNP, SPG57, and TRKT3.
- Plays a role in the normal dynamic function of the endoplasmic reticulum (ER) and its associated microtubules (PubMed:23479643, PubMed:27813252). Related references: PMID:23479643 PMID:27813252
- Required for secretory cargo traffic from the endoplasmic reticulum to the Golgi apparatus (PubMed:21478858). Related references: PMID:21478858
- Subcellularly localized to the endoplasmic reticulum membrane, where it maintains ER morphology and facilitates interactions with the microtubule cytoskeleton.
- Subject to post-translational modifications including phosphorylation, acetylation, and methylation, which may regulate its function in membrane trafficking.
- Mutations in TFG are associated with autosomal recessive hereditary spastic paraplegia (SPG57) and hereditary motor and sensory neuropathy with proximal dominance (HMSNP), linking it to neurodegenerative diseases.
- Chromosomal rearrangements leading to TFG fusion proteins act as proto-oncogenic drivers in certain cancers, such as anaplastic large cell lymphoma and papillary thyroid carcinoma.

- The immunogen is a synthetic peptide spanning residues 301–400 of human TFG; confirm detection of the full-length 43 kDa protein under your experimental conditions.
- For Western blot (WB), consider using standard protocols and include positive controls expressing TFG at the expected molecular weight.
- Cross-reactivity with mouse has been reported; validate in murine samples if your study involves mouse models, and optimize antibody dilution accordingly.
