Biological Background: TRIM69 Function and Localization

  • TRIM69 (also known as Trif, HSD34, RNF36, or HSD-34) is an E3 ubiquitin-protein ligase belonging to the tripartite motif (TRIM) family, characterized by a RING finger domain, B-box, and coiled-coil region.
  • Plays an important role in antiviral immunity by ubiquitinating viral proteins such as dengue virus NS3, thereby restricting infection by dengue virus and vesicular stomatitis Indiana virus. Related references: PMID:23131556, PMID:30142214, PMID:31375575, PMID:30844644
  • Acts as a key mediator of type I interferon-induced microtubule stabilization by directly associating with microtubules, independent of its E3 ligase activity. Related references: PMID:36251989
  • Contributes to cataract formation together with TP53 by inhibiting UVB-induced cell apoptosis and reactive oxygen species (ROS) production through ubiquitination of TP53. Related references: PMID:30844644
  • Regulates centrosome dynamics and mitotic progression by ubiquitinating STK3/MST2, leading to its redistribution to the perinuclear cytoskeleton and subsequent phosphorylation by PLK1. Related references: PMID:37739411
  • Localizes to the cytoplasm, nucleus, nuclear speckles, and the microtubule organizing center (centrosome).
  • Contains a RING-type zinc finger and coiled-coil domain; undergoes alternative splicing and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1–341 at the N-terminal region of human TRIM69; consider using a positive control such as overexpressed TRIM69 or known expressing cell lines (e.g., HeLa, HEK293) when establishing assays.
  • For Western blotting, a band around 57 kDa is expected; validate in the relevant sample system, and optimize antibody dilution based on target abundance.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization conditions may be required; signal localization should mirror known subcellular distribution (cytoplasm, nucleus, centrosome).
  • This antibody is reported to cross-react with mouse TRIM69; species-specific validation is recommended if used outside of human and mouse.
