Biological Background: TRIM59 Function and Localization
- TRIM59 (Tripartite motif-containing protein 59), also referred to as RING finger protein 104 (RNF104) or tumor suppressor TSBF-1, is a member of the TRIM family of E3 ubiquitin ligases involved in diverse cellular processes including development and innate immunity. - It functions as a negative regulator of innate immune signaling by suppressing RIG-I-like receptor (RLR)-induced activation of interferon regulatory factors IRF3 and IRF7, as well as NF-κB, through interaction with the adaptor protein ECSIT. Related references: PMID:22588174
- TRIM59 modulates autophagy by regulating BECN1 at both the transcriptional and post-translational levels; it downregulates BECN1 transcription via NF-κB pathway inhibition and controls TRAF6-mediated Lys-63-linked ubiquitination of BECN1, impacting the BECN1-PIK3C3 complex assembly.
