Biological Background: TRIM45 Function and Localization

  • TRIM45, also known as RNF99, is an E3 ubiquitin-protein ligase that belongs to the tripartite motif (TRIM) family, featuring a RING finger domain, B-box, and coiled-coil region. It is involved in the regulation of inflammatory responses.
  • TRIM45 mediates 'Lys-48'-linked polyubiquitination of TAB2, a regulatory protein of the kinase TAK1, targeting it for proteasomal degradation. This process inhibits TLR-mediated inflammatory immune signaling.
    Related references: PMID:36681779
  • May also function as a transcriptional repressor in the mitogen-activated protein kinase (MAPK) signaling pathway.
  • Localizes to both the cytoplasm and nucleus, consistent with its roles in signal transduction and transcriptional regulation.
  • Expressed in multiple tissues, including skeletal muscle, brain, heart, and pancreas.
  • Exhibits alternative splicing, producing multiple isoforms that may have distinct functional properties.
  • The protein contains zinc-finger motifs (including the RING domain), a coiled-coil region, and undergoes post-translational modifications such as ubiquitination, which are critical for its ligase activity and substrate interaction.
  • With a predicted molecular weight of approximately 64 kDa (64359 Da), TRIM45 is subject to proteasomal degradation itself and is identified in proteomic studies.

Experimental Guidance and Technical Tips

  • This polyclonal antibody was raised against a recombinant protein covering amino acids 100–400 of human TRIM45, a region likely encompassing functional domains; it can be used for immunofluorescence/immunocytochemistry (IF/ICC) and ELISA applications.
  • For IF/ICC, optimize fixation and permeabilization protocols to detect TRIM45 in both cytoplasmic and nuclear compartments. Consider using appropriate isotype controls.
  • In ELISA, validate the antibody as a capture or detection reagent in the specific sample matrix (e.g., cell lysates, tissue extracts) to ensure sensitivity and specificity.
  • The antibody cross-reacts with mouse TRIM45, enabling its use in mouse model studies; however, verify cross-reactivity in your experimental setup.
  • As a polyclonal antibody, lot-to-lot variability may occur; new lots should be validated for consistency in your assays.
