Biological Background: TRIM41 Function and Localization
- E3 ubiquitin-protein ligase TRIM41 (also known as RINCK) is a member of the tripartite motif (TRIM) family, characterized by a RING finger domain, B-box, and coiled-coil regions.
- Functions as an E3 ligase essential for innate antiviral response by directly targeting influenza A virus and vesicular stomatitis virus nucleoproteins for ubiquitination and proteasomal degradation, thereby limiting viral infections. Related references: PMID:28169297, PMID:29899090, PMID:31979016
- Activates the innate antiviral response by catalyzing monoubiquitination of CGAS, thereby activating CGAS. Related references: PMID:29760876
- Mediates 'Lys-63'-linked polyubiquitylation of BCL10, which in turn hubs NEMO for activation of NF-kappa-B and IRF3 pathways (By similarity).
- Catalyzes ubiquitin-mediated degradation of various substrates including protein kinase C, ZSCAN21, and TOP3B, suggesting roles beyond immune response. Related references: PMID:17893151, PMID:33378676
- Localizes to both the cytoplasm and nucleus.
- Expressed in multiple tissues with highest levels in heart and skeletal muscle.
- Contains a RING-type zinc finger and is classified under phosphoprotein, transferase, and Ubl conjugation pathway keywords; undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 421-630 of human TRIM41; for Western blotting, consider using appropriate loading controls and validating the band size (predicted ~72 kDa) in the relevant sample system.
- As a polyclonal antibody, it may recognize multiple epitopes, which can enhance detection sensitivity but may also increase background; optimize blocking and washing conditions.
- For ELISA applications, consider titrating the antibody and confirming specificity with recombinant protein or peptide competition assays.
- Cross-reactivity with mouse and rat is reported; perform species-specific validation if switching between model organisms.
