Biological Background: TRIM39 Function and Localization
- TRIM39 is a member of the tripartite motif (TRIM) family, characterized by a RING finger domain, B-box, and coiled-coil region.
- It functions as an E3 ubiquitin-protein ligase that may facilitate apoptosis by inhibiting APC/C-Cdh1-mediated ubiquitination and degradation of the pro-apoptotic protein MOAP1. Related references: PMID:19100260, PMID:22529100
- TRIM39 regulates the G1/S transition and DNA damage-induced G2 arrest by stabilizing CDKN1A/p21 via competition with DTL for binding, thus preventing its ubiquitination and degradation. Related references: PMID:23213251
- It negatively regulates the canonical NF-kappa-B signaling pathway by stabilizing CACTIN in an ubiquitination-independent manner. Related references: PMID:26363554
- Subcellularly, TRIM39 localizes to the cytoplasm, cytosol, mitochondrion, and nucleus.
- It is ubiquitously expressed, with high levels in brain, heart, kidney, liver, skeletal muscle, spleen, and testis.
- The protein contains a zinc-finger domain, coiled-coil region, and is involved in the ubiquitin conjugation pathway.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide within amino acids 350–450 of human TRIM39. Consider that it may recognize various isoforms or post-translationally modified forms.
- For Western blot, a band around 60 kDa is expected; however, minor bands might appear due to alternative splicing or modifications. Validate in your sample system.
- Standard ELISA protocols can be employed; optimize coating and detection conditions based on your specific assay requirements.
- Expression levels vary across tissues, so include appropriate positive and negative controls.
