Biological Background: TRIM24 Function and Localization
- TRIM24 (Transcription intermediary factor 1-alpha) is a member of the tripartite motif (TRIM) family, also known as TIF1α, RNF82, and PTC6.
- Acts as a transcriptional coactivator that interacts with various nuclear receptors, including androgen receptor (AR), glucocorticoid receptor (GCR/NR3C1), thyroid hormone receptor (TR), estrogen receptor alpha (ESR1), and retinoic acid receptors (RARA), thereby up-regulating ligand-dependent transcription and modulating hepatocyte proliferation (By similarity).
- Interacts with chromatin, demonstrating highest affinity for histone H3 unmodified at Lys-4 (H3K4me0) and acetylated at Lys-23 (H3K23ac), indicating its role in chromatin-mediated transcription regulation.
- Functions as an E3 ubiquitin-protein ligase and participates in the DNA damage response by forming an autoregulatory feedback loop with TP53: ATM kinase phosphorylates TRIM24, leading to its ubiquitination and degradation, while TRIM24 subsequently ubiquitinates TP53 for degradation, thus controlling cell proliferation and apoptosis.
