Biological Background: TRIM24 Function and Localization

  • TRIM24 (Transcription intermediary factor 1-alpha) is a member of the tripartite motif (TRIM) family, also known as E3 ubiquitin-protein ligase TRIM24, RING finger protein 82, and TIF1-alpha. It functions as a transcriptional coactivator and an E3 ubiquitin ligase.
  • TRIM24 interacts with chromatin by binding histone H3 when it is unmodified at Lys-4 (H3K4me0) and acetylated at Lys-23 (H3K23ac), mediating gene regulation. Its PHD finger and bromodomain recognize these modifications.
  • Early in the DNA damage response, ATM kinase phosphorylates TRIM24, leading to its ubiquitination and degradation. After sufficient DNA repair, TP53 activates TRIM24 transcription, and TRIM24 in turn ubiquitinates TP53 for degradation, forming an autoregulatory feedback loop. Related references: PMID:24820418

  • This antibody is suitable for ELISA. Due to its polyclonal nature, we recommend including a no-primary antibody control and pre-immune serum control when setting up assays.
