Biological Background: TRIM17 Function and Localization
- TRIM17 (tripartite motif-containing 17), also known as RNF16, TERF, or RBCC, is a member of the TRIM/RBCC family bearing a RING finger, B-box, and coiled-coil domains.
- Acts as an E3 ubiquitin ligase that plays important roles in the regulation of neuronal apoptosis, selective autophagy, and cell proliferation. Related references: PMID:19358823 PMID:22023800 PMID:27562068
- Stimulates proteasome-dependent degradation of the kinetochore protein ZWINT, thereby negatively regulating cell proliferation.
- Inhibits autophagic degradation by assembling MCL1 into complexes with BECN1, and is required for midbody autophagy.
- Controls neuronal apoptosis through ubiquitination and degradation of MCL1.
- Localizes to the cytoplasm and lysosomes.
- Almost exclusively expressed in testis.
- The protein bears metal-binding zinc finger motifs, consistent with its transferase activity, and undergoes alternative splicing to produce multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 280-477, covering the C‑terminal region including the B30.2/SPRY domain and coiled‑coil motif; consider that the antibody may detect isoforms retaining this region.
- For Western blot, the predicted molecular weight is ~54 kDa; validate with appropriate positive controls (e.g., testis lysates) and confirm specificity in your sample system.
- In ELISA applications, ensure the coating antigen matches the immunogen region for optimal detection.
- Reported cross‑reactivity with mouse and rat orthologs should be verified experimentally in each species.
- Given the testis‑specific expression, consider using testis‑derived samples for expression studies.
