  • TRIB3 (Tribbles homolog 3) is an inactive protein kinase that acts as a key regulator of the integrated stress response (ISR), a cellular adaptation to various stresses.   • Functions as a negative feedback regulator of ATF4-dependent transcription: TRIB3 expression is induced by ATF4, and the protein then binds ATF4 to suppress its transcriptional activity, fine-tuning the ISR.
  • Disrupts insulin signaling by directly binding to Akt kinases and blocking their activation, potentially by masking the Thr-308 phosphorylation site in AKT1.
