Biological Background: TRIB3 Function and Localization
- TRIB3 (Tribbles homolog 3) is an inactive protein kinase that acts as a key regulator of the integrated stress response (ISR), a cellular adaptation to various stresses. - Functions as a negative feedback regulator of ATF4-dependent transcription: TRIB3 expression is induced by ATF4, and the protein then binds ATF4 to suppress its transcriptional activity, fine-tuning the ISR.
- Disrupts insulin signaling by directly binding to Akt kinases and blocking their activation, potentially by masking the Thr-308 phosphorylation site in AKT1.
- Interacts with the NF-kappa-B transactivator p65/RELA, inhibiting its phosphorylation and transcriptional activation, thereby modulating inflammatory and survival pathways. Related references: PMID:12736262

- The immunogen is a synthetic peptide spanning the N-terminal region (amino acids 1-100) of human TRIB3, making it suitable for detecting the full-length protein in western blotting and for peptide-based ELISA formats.
- A band of approximately 40 kDa is expected in western blot. Optimal antibody concentrations should be determined empirically; a starting dilution of 1:1000–1:5000 is often suitable, and validation with appropriate positive and negative controls is recommended.
- For ELISA, ensure the peptide epitope is accessible; both direct and sandwich formats may be explored depending on the assay design.
