Biological Background: TRH Function and Localization

• Pro-thyrotropin-releasing hormone (pro-TRH), also known as prothyroliberin, is a secreted precursor protein that undergoes post-translational processing to generate mature TRH peptides.
• It is a key component of the hypothalamic-pituitary-thyroid axis, controlling the secretion of thyroid-stimulating hormone (TSH) and regulating thyroid hormone synthesis.
• Beyond its endocrine role, pro-TRH functions as a modulator of hair growth; it promotes hair-shaft elongation, prolongs the anagen (growth) phase, and antagonizes catagen (termination) through TGFB2 signaling.
• Pro-TRH stimulates proliferation and inhibits apoptosis of hair matrix keratinocytes, contributing to hair follicle cycling.
