Biological Background: TREX1 Function and Localization
- TREX1 is a major cellular 3'-to-5' DNA exonuclease that digests single-stranded DNA (ssDNA) and double-stranded DNA (dsDNA) with mismatched 3' termini, preventing cell-intrinsic initiation of autoimmunity by metabolizing DNA fragments from endogenous retroelements including L1, LTR and SINE elements. - Upon genome instability, TREX1 is recruited from the endoplasmic reticulum membrane to ruptured micronuclei, where it degrades micronuclear DNA to limit cGAS activation and subsequent type I interferon production, thereby preventing autoinflammatory responses. Related references: PMID:33476576
- TREX1 processes ssDNA polynucleotide species arising from the processing of aberrant DNA replication intermediates, preventing chronic ATM-dependent checkpoint activation. Related references: PMID:18045533

Experimental Guidance and Technical Tips
- For Western blot (WB), use whole cell lysates from human, mouse, or rat tissues known to express TREX1. The predicted molecular weight is ~33 kDa; validate with positive and negative controls.
- For immunofluorescence (IF/ICC), optimize fixation and permeabilization protocols to detect TREX1 in its nuclear and cytoplasmic compartments. Consider co-localization with organelle markers.
