- Heat shock protein 75 kDa, mitochondrial (TRAP1/HSP75) is a member of the heat shock protein 90 family functioning as an ATP-dependent molecular chaperone.
- It plays a critical role in maintaining mitochondrial integrity and polarization, acting downstream of PINK1 and mitochondrial complex I.
- TRAP1 negatively regulates mitochondrial respiration, modulating the balance between oxidative phosphorylation and aerobic glycolysis, possibly through inhibition of succinate dehydrogenase (SDHA) and modulation of mitochondrial SRC.
- The protein localizes predominantly to the mitochondrial inner membrane and matrix, consistent with its role in mitochondrial homeostasis.
- Tissue expression is broad, including skeletal muscle, liver, heart, brain, kidney, pancreas, lung, placenta, and bladder; expression levels are reduced in bladder cancer and renal cell carcinoma but elevated in several other tumor types.
