Biological Background: TRAP1 Function and Localization
- TRAP1, also known as Heat shock protein 75 kDa, mitochondrial, Hsp75, HSP90L, or TNFR-associated protein 1, is a mitochondrial chaperone with ATPase activity.
- It maintains mitochondrial function and polarization downstream of PINK1 and mitochondrial complex I, acting as a negative regulator of mitochondrial respiration to modulate the balance between oxidative phosphorylation and aerobic glycolysis.
- TRAP1 belongs to the heat shock protein 90 family and is encoded by the TRAP1 gene (also known as HSP75, HSPC5) on chromosome 16.
- It localizes to the mitochondrion, predominantly in the inner membrane and matrix.
- Post-translational modifications include acetylation and phosphorylation, as identified by proteomics.
- TRAP1 expression is found in various tissues including skeletal muscle, liver, heart, brain, kidney, pancreas, lung, placenta, and bladder, with altered expression in certain cancers such as reduced levels in bladder and renal cell carcinoma but increased in other tumors.
- The protein has a molecular weight of approximately 80 kDa (calculated 80,110 Da) and may exist in multiple isoforms due to alternative splicing.
