Biological Background: Translin Function and Localization
- Translin, also known as Component 3 of promoter of RISC (C3PO), is a DNA/RNA-binding protein encoded by the TSN gene.
- As a DNA-binding protein, it specifically recognizes consensus sequences at breakpoint junctions in chromosomal translocations, mostly involving immunoglobulin (Ig) and T-cell receptor gene segments, and binds single-stranded DNA ends generated during recombination.
- Exhibits both single-stranded and double-stranded endoribonuclease activity, and is implicated in RNA-induced silencing complex (RISC) activation by facilitating endonucleolytic cleavage of the siRNA passenger strand.
- Localizes to both the cytoplasm and nucleus, consistent with its roles in DNA repair/recombination and RNA processing.
- Protein weight is approximately 26 kDa; the antibody detects a band around 26 kDa in WB.
- Post-translational modifications include acetylation and phosphorylation; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 50–150 of human Translin (NP_004613.1), a region highly conserved across human, mouse, and rat, supporting the observed cross-reactivity. For species not listed, verify sequence homology before use.
- For Western blot (WB), expect a specific band at ~26 kDa. Optimize blocking and antibody dilution for your sample type.
- For IHC-P and IF/ICC, consider the cytoplasmic and nuclear localization; appropriate fixation and permeabilization protocols are recommended.
- This polyclonal antibody is suitable for ELISA; titrate the antibody to achieve an optimal signal-to-noise ratio.
