Biological Background: Transglutaminase 2 Function and Localization
- Also known as tissue transglutaminase (tTG) and TG2, TGM2 is a calcium-dependent acyltransferase that catalyzes protein cross-linking, amine incorporation, and deamidation. Related references: PMID:23941696, PMID:31991788, PMID:9252372
- It plays a key role in apoptosis by cross-linking cytoskeletal and extracellular matrix proteins to stabilize dying cells and prevent leakage of intracellular components. Related references: PMID:7935379, PMID:9252372
- TGM2 modifies histone H3 via serotonylation at Gln-5 (H3Q5ser) during neuronal differentiation, facilitating transcription. Related references: PMID:30867594
- In dopaminergic neurons, TGM2 catalyzes dopaminylation of histone H3 at Gln-5 (H3Q5dop), regulating relapse-related transcriptional plasticity. Related references: PMID:32273471
- The enzyme is primarily cytosolic but also localizes to the nucleus, chromosomes, mitochondria, cell membrane, and is secreted into the extracellular space.
- Its cross-linking activity is inhibited by GTP under physiological conditions; calcium binding relieves this inhibition in response to stress. Related references: PMID:18092889, PMID:7592956, PMID:7649299
- TGM2 deamidates specific glutamine residues, such as in dietary gliadin, a process linked to celiac disease pathogenesis. Related references: PMID:9623982
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 299-548) within the catalytic core; consider validating with appropriate positive and negative controls in your sample system.
- For WB, the predicted molecular weight is 77 kDa, but post-translational modifications may cause electrophoretic shifts; include suitable molecular weight markers.
- In IHC-P, ensure optimal antigen retrieval, as TGM2 may be cross-linked in fixed tissues.
- For ELISA, we recommend titrating the coating antigen and antibody to establish optimal signal-to-noise ratios.
- Given confirmed cross-reactivity with mouse and rat, this antibody may be compatible with rodent models; validate in the relevant species.
