  • Transgelin, also known as SM22-alpha, WS3-10, or 22 kDa actin-binding protein, is encoded by the TAGLN gene and is a member of the calponin family of actin-binding proteins.
  • It functions as an actin cross-linking and gelling protein, thereby regulating actin filament dynamics and cytoskeletal organization.
  • Transgelin is involved in calcium-dependent interactions and contributes to the contractile properties of the cell, which may play a role in replicative senescence.
  • The protein predominantly localizes to the cytoplasm.
  • It undergoes multiple post-translational modifications, including acetylation, methylation, and phosphorylation.
  • Transgelin is a marker of smooth muscle differentiation and is also expressed in other cell types where it participates in actin remodeling.

Experimental Guidance and Technical Tips

  • The immunogen was designed from a synthetic peptide spanning amino acids 100-201 of human TAGLN (NP_003177.2). This region shows high conservation among human, mouse, and rat, consistent with the observed cross-reactivity.
  • For western blot (WB), a specific band around 23 kDa is expected. Endogenous transgelin expression levels may vary across cell and tissue types.
  • In immunofluorescence (IF-P) and immunohistochemistry (IHC-P), consider optimizing permeabilization and antigen retrieval conditions for the target sample.
  • ELISA can be employed for quantitative detection; appropriate blocking and washing steps should be validated.
  • As a recombinant rabbit monoclonal antibody (Rabbit PolymAb®), high lot-to-lot consistency is expected, but experimental conditions should be titrated individually.
