  • TRAFD1 (FLN29) is a TRAF-type zinc finger domain-containing protein encoded by the TRAFD1 gene (Gene ID: 10906) in humans.
  • It acts as a negative feedback regulator that controls excessive innate immune responses by modulating TLR4 and RLH pathways.
  • The protein directly interacts with TRAF6, inhibiting TLR4-mediated NF-kappa-B activation (By similarity).
  • In the RLH pathway, TRAFD1 negatively regulates signaling downstream of MAVS and upstream of NF-kappa-B and IRF3 (By similarity).
  • Zinc finger motifs enable metal binding and are critical for protein structure and function; TRAFD1 is also subject to acetylation and phosphorylation.
  • Alternative splicing generates multiple isoforms, expanding the functional repertoire of TRAFD1 in immune regulation.
