Biological Background: TRAFD1 Function and Localization
- TRAFD1 (FLN29) is a TRAF-type zinc finger domain-containing protein encoded by the TRAFD1 gene (Gene ID: 10906) in humans.
- It acts as a negative feedback regulator that controls excessive innate immune responses by modulating TLR4 and RLH pathways.
- The protein directly interacts with TRAF6, inhibiting TLR4-mediated NF-kappa-B activation (By similarity).
- In the RLH pathway, TRAFD1 negatively regulates signaling downstream of MAVS and upstream of NF-kappa-B and IRF3 (By similarity).
- Zinc finger motifs enable metal binding and are critical for protein structure and function; TRAFD1 is also subject to acetylation and phosphorylation.
- Alternative splicing generates multiple isoforms, expanding the functional repertoire of TRAFD1 in immune regulation.
Experimental Guidance and Technical Tips
- The immunogen used corresponds to the N-terminal region (aa 1-260) of human TRAFD1, which includes part of the zinc finger domain; thus, the antibody may recognize multiple isoforms sharing this region.
- For Western blot (WB), a band at approximately 65 kDa is expected in human samples; consider using positive controls such as immune-stimulated cell lysates where TRAFD1 expression may be upregulated.
- In ELISA, the antibody can be used for direct or indirect detection of recombinant or endogenous TRAFD1; validate coating and detection conditions for your specific assay format.
